The eight children, three women and five men died when a blast blew apart their minibus.
A police spokesman in Kandahar said the vehicle had been destroyed as it drove through Arghandab district to the Shah Agha shrine.
There were no survivors among the family, from Sarwan Qala in neighbouring Helmand province.
In May, 368 Afghan civilians killed in the conflict, and nearly 600 wounded, in what the UN said was the highest monthly total since its records began four years ago.
Other fatal attacks on Saturday included the assassination of a senior police official in a suicide bombing in Khost that also killed three bystanders.
The true toll was probably higher because much of the fighting is in inaccessible areas where deaths go unrecorded.
The United Nations estimated 45 of May's deaths were caused by Nato or Afghan forces and 301 by insurgents.
Georgette Gagnon, director of human rights for the UN in Afghanistan, said: "We are very concerned that civilian suffering will increase even more over the summer fighting season which historically brings the highest numbers of civilian casualties." Homemade Taliban bombs dug into roads, tracks and buildings and designed to blow up passing Nato patrols or Afghan government targets accounted for two-fifths of the dead.
The crude devices often detonate indiscriminately and such a bomb was blamed for the destruction of the minibus.
The Taliban high command has apparently issued directives ordering fighters to avoid civilian casualties, but human rights groups have said the death toll from insurgents continued to rise.
May's toll also included at least nine dead from a bungled Nato helicopter attack in Naw Zad district of Helmand which prompted Hamid Karzai to demand an end to coalition air strikes.
Violence is again at record levels in Afghanistan and is expected to mount sharply in the next few weeks as Taliban finish harvesting opium and begin their summer offensive.
